چکیده

Fatty acid and alkane hydroxylation by the inducible, cyanide-sensitive enzyme system of Pseudomonas oleovorans involves three enzymes : NADH-rubredoxin reductase, rubredoxin, and an w-hydroxylase. The w-hydroxylase is a yellow protein containing one atom of iron per polypeptide chain of molecular weight 42,000. Catalytic activity is lost upon removal of the iron and is restored when ferrous ions are incubated with the apohydroxylase. Hydroxylase preparations from which most of the phospholipid has been removed exhibit decreased activity unless supplemented with a phospholipid fraction from this bacterium or with dilauroylglyceryl3-phosphorylchol ine.

Differences and Comparisons of the Properties and Reactivities of Iron(III)–hydroperoxo Complexes with Saturated Coordination Sphere

Heme and nonheme monoxygenases and dioxygenases catalyze important oxygen atom transfer reactions to substrates in the body. It is now well established that the cytochrome P450 enzymes react through the formation of a high-valent iron(IV)-oxo heme cation radical. Its precursor in the catalytic cycle, the iron(III)-hydroperoxo complex, was tested for catalytic activity and found to be a sluggish...

Oxidative phosphorylation in fractionated bacterial systems. XXIX. The involvement of nonheme iron in the respiratory pathways of Mycobacterium phlei.

Spectrophotometric evidence for the reduction of nonheme iron in the respiratory particles of Mycobacferium phlei was obtained by trapping the metal as ferrous o-phenanthrolinate. The protein-bound iron-o-phenanthroline complex had an absorption maximum at 510 mp, and the extent of reduction of nonheme iron was followed by observing the increase in absorbance at 510 to 540 rnp on the addition o...

Regioselectivity of aliphatic versus aromatic hydroxylation by a nonheme iron(II)-superoxo complex.

Many enzymes in nature utilize molecular oxygen on an iron center for the catalysis of substrate hydroxylation. In recent years, great progress has been made in understanding the function and properties of iron(IV)-oxo complexes; however, little is known about the reactivity of iron(II)-superoxo intermediates in substrate activation. It has been proposed recently that iron(II)-superoxo intermed...

Purification and characterization of phthalate oxygenase and phthalate oxygenase reductase from Pseudomonas cepacia.

An enzymatic system has been isolated that catalyzes dihydroxylation of phthalate to form 1,2-dihydroxy-4,5-dicarboxy-3,5-cyclohexadiene with consumption of NADH and O2. This system is comprised of two proteins: a flavo-iron-sulfur protein with NADH-dependent oxidoreductase activity and a nonheme iron protein with oxygenase activity. Phthalate oxygenase is a large (approximately 217 kDa) protei...

Nonheme iron(II) complexes of macrocyclic ligands in the generation of oxoiron(IV) complexes and the catalytic epoxidation of olefins.

Mononuclear nonheme oxoiron(IV) complexes bearing 15-membered macrocyclic ligands were generated from the reactions of their corresponding iron(II) complexes and iodosylbenzene (PhIO) in CH(3)CN. The oxoiron(IV) species were characterized with various spectroscopic techniques such as UV-vis spectrophotometer, electron paramagnetic resonance, electrospray ionization mass spectrometer, and resona...
